The Sample Peak Universal Router (SPUR) is a simple electrophoretic separation device designed for isolation of individual components from a mixture and sequential peak concentrating prior to dispensing for further manipulation or analysis. SPUR operates on the basis of balancing molecular velocities imposed by a chromatographic flow and electromigration, resulting in discrete bands of sample. The chromatographic velocity is simply generated by flow through the separation chamber, pushing sample components towards the exit of the chamber. The opposing electromigration results from a gradient in an electric field using a chamber of a pre-defined geometry. In most instances, a linear field gradient is used for focusing sample bands. SPUR separation of products is a biocompatible process with use of buffers of moderate salt concentrations and pHs. |
